Overview

This investigational cell therapy, developed by the National Heart, Lung, and Blood Institute (NHLBI), involves the co-infusion of CD34+ selected cells from a haploidentical related donor and a single unrelated umbilical cord blood (UCB) unit. It is designed for patients with severe aplastic anemia (SAA) or myelodysplastic syndrome (MDS) who lack a matched donor and are refractory to immunosuppressive therapy. The haploidentical CD34+ cells are T-cell depleted using the Miltenyi CliniMacs system and provide rapid, transient myeloid engraftment (typically within 10 days) to bridge the period of neutropenia. Long-term, durable hematopoiesis is then established by the umbilical cord blood unit. This 'haplo-cord' approach aims to mitigate the high rates of graft failure and delayed recovery associated with UCB-only transplants in SAA.
