Overview

This investigational cellular immunotherapy consists of haploidentical donor natural killer (NK) cells activated with recombinant human interleukin-15 (rhIL-15). Developed by researchers at the Masonic Cancer Center, University of Minnesota, the therapy is designed to treat patients with relapsed or refractory acute myeloid leukemia (AML). The treatment protocol involves the collection of NK cells from a partially HLA-matched (haploidentical) related donor, followed by ex vivo depletion of T and B lymphocytes (CD3/CD19 depletion) to reduce the risk of graft-versus-host disease. The enriched NK cells are activated overnight with IL-15 before being infused into the patient. To enhance the in vivo expansion and persistence of these donor cells, patients receive a lymphodepleting preparative regimen (cyclophosphamide and fludarabine) followed by systemic administration of rhIL-15, either intravenously or subcutaneously.
