Haploidentical stem cells + natural killer (NK) cells refers to a combination cellular therapy in which hematopoietic stem cells from a haploidentical (half-matched, usually familial) donor are transplanted into a patient, often for the treatment of hematologic malignancies such as acute myeloid leukemia (AML), and are combined with an infusion of donor-derived NK cells. The rationale is that alloreactive NK cells can mediate antileukemic effects, enhance engraftment, and reduce relapse risk and graft-versus-host disease (GVHD). In this approach, patients typically receive cytoreductive or conditioning chemotherapy followed by sequential infusions of ex-vivo expanded or activated haploidentical NK cells prior to or after the stem cell transplant. This strategy is under investigation primarily for refractory AML and other high-risk blood cancers[1][2][3].
