HB-201 + pembrolizumab is a combination therapy under investigation for the treatment of cancers associated with human papillomavirus (HPV), particularly HPV16-positive tumors. HB-201 is a replication-competent, live attenuated arenavirus-based cancer immunotherapy vector developed by Hookipa Pharma. It encodes a non-oncogenic fusion protein of HPV16 E7 and E6 antigens, designed to stimulate an immune response specifically against HPV16-expressing tumor cells by inducing cytotoxic T lymphocytes[6][8]. Pembrolizumab is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD‑1) receptor on T-cells, blocking its interaction with PD-L1/PD-L2 ligands and thereby enhancing anti-tumor immune activity[1][5]. The combination aims to leverage both direct antigen-specific immune activation (via HB‑201) and checkpoint inhibition (via pembrolizumab) for synergistic anti-cancer effects.
