HB0025 + pemetrexed + carboplatin is an investigational combination regimen being studied primarily in advanced non-squamous non-small cell lung cancer (NSCLC). HB0025 is a recombinant fusion protein targeting the programmed cell death protein 1 (PD-1) immune checkpoint, acting as an immunotherapeutic agent. Pemetrexed is an antifolate chemotherapy agent that targets folate-dependent enzymes, interfering with DNA and RNA synthesis, and carboplatin is a platinum-based chemotherapeutic that forms DNA crosslinks, leading to apoptosis. This combination is designed to maximize cancer cell killing through both immune modulation (via HB0025) and cytotoxic approaches (via pemetrexed and carboplatin). The regimen is being evaluated for safety, tolerability, and efficacy in Phase Ib/II clinical trials for patients with recurrent, metastatic, or locally advanced non-squamous NSCLC who have not received prior systemic therapy and do not carry EGFR sensitive mutations or ALK fusion gene sensitive mutations[3].
