This is a combination of HCB101, a novel Fc-fusion protein engineered from the IgV domain of human signal regulatory protein alpha (SIRPα) designed to selectively block the CD47-SIRPα pathway, and cetuximab, a recombinant chimeric monoclonal antibody that targets the epidermal growth factor receptor (EGFR). HCB101 promotes macrophage-mediated phagocytosis of tumor cells by disrupting the "don't eat me" signal mediated via CD47, activating innate and adaptive immunity with reduced red blood cell binding, minimizing hematologic toxicity. Cetuximab inhibits EGFR-associated signaling, causing inhibition of tumor growth, induction of apoptosis, and disruption of angiogenesis. This combination is investigated mainly for advanced solid tumors, including metastatic colorectal cancer and squamous cell carcinoma of the head and neck, aiming to enhance anti-tumor immunity and cell killing via dual targeting of immune evasion (CD47) and growth signaling (EGFR)[1][2][3][4][7][9][10].
