Overview

HDM201 + MBG453 is an experimental combination therapy consisting of HDM201 (also known as siremadlin), an oral small molecule inhibitor of the HDM2-p53 interaction, and MBG453 (also known as sabatolimab), a monoclonal antibody targeting T-cell immunoglobulin and mucin-domain containing-3 (TIM-3). HDM201 is designed to reactivate the p53 tumor suppressor pathway by inhibiting the negative regulator HDM2, thereby promoting apoptosis in tumor cells with wild-type TP53. MBG453 inhibits the TIM-3 immune checkpoint, potentially enhancing anti-tumor immune responses. This combination is being investigated for its safety and antitumor activity in acute myeloid leukemia (AML) and high-risk myelodysplastic syndrome (MDS), particularly in cases with wild-type TP53, by targeting both intrinsic tumor cell death and extrinsic immune-mediated mechanisms[1][3].
