Hemay022 is a novel, orally available small-molecule and irreversible tyrosine kinase inhibitor targeting both epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 2 (HER2). It covalently binds to and inhibits EGFR/HER2, blocking downstream signaling pathways involved in tumor cell proliferation. Developed by Tianjin Hemay Biotech, Hemay022 has demonstrated anti-tumor activity in preclinical models and is being investigated for HER2-positive advanced breast cancer[1][2][5][7]. Exemestane is an oral steroidal aromatase inhibitor that irreversibly binds to the aromatase enzyme, preventing the conversion of androgens into estrogen. This mechanism reduces estrogen levels in postmenopausal women, inhibiting the growth of hormone-sensitive breast cancer cells[6][8]. The combination of Hemay022 with an aromatase inhibitor such as exemestane is under clinical investigation for enhanced efficacy in treating advanced or metastatic breast cancer.
