The HER2 peptide vaccine developed by the Technical University of Munich is an investigational peptide-based cancer vaccine designed to treat solid tumors, specifically metastasized breast and gastric cancers, that exhibit moderate expression of the HER2/neu (ERBB2) protein. The vaccine consists of specific peptides derived from the HER2/neu protein that are intended to stimulate a cytotoxic T-lymphocyte (CTL) response against tumor cells. In clinical protocols (such as NCT01570036), the vaccine is administered as part of a multi-component regimen: it is combined with sargramostim (GM-CSF) as an immunological adjuvant and topical imiquimod (Aldara) to enhance the local immune response. Additionally, patients receive low-dose cyclophosphamide (Endoxan) as a pre-treatment to deplete regulatory T cells (Tregs), thereby reducing immune suppression and potentially increasing the vaccine's efficacy.
