HER2-specific chimeric antigen receptor T cells + CAdVEC is an investigational combination immuno-oncology regimen developed by Baylor College of Medicine for advanced HER2-positive solid tumors. The therapy combines autologous T cells genetically engineered to express a chimeric antigen receptor targeting human epidermal growth factor receptor 2 with CAdVEC, a binary oncolytic and helper-dependent adenoviral platform intended to remodel the tumor microenvironment. The CAR-T component is designed to recognize and kill HER2-expressing tumor cells, while the adenoviral component is intended to enhance local immune activation, improve CAR-T-cell trafficking and persistence, and augment antitumor activity in solid tumors where adoptive cell therapy alone may be less effective.
