Overview

Herzuma + gedatolisib is a combination therapy consisting of Herzuma (a trastuzumab biosimilar) and gedatolisib (a dual PI3K/mTOR inhibitor). This combination targets HER2-positive metastatic breast cancer, particularly in patients with PIK3CA aberrations who have progressed after multiple lines of HER2-directed therapies. Herzuma binds to the HER2/neu receptor, reducing HER2 expression and inhibiting tumor-supporting blood vessel growth. Gedatolisib targets the mTOR/PI3K signaling pathway, which plays a crucial role in tumor growth and proliferation. The combination has shown promising results in clinical trials, with an overall response rate of 58.8% and a disease control rate of 82.4% in patients with HER2-positive metastatic breast cancer with PIK3CA aberrations.
