HFO-1234ze(E), also known as trans-1,3,3,3-tetrafluoropropene, is a next-generation hydrofluoroolefin (HFO) propellant developed by AstraZeneca in collaboration with Honeywell. It is designed for use in pressurized metered dose inhalers (pMDIs) to replace current hydrofluoroalkane (HFA) propellants like HFA-134a, which have high global warming potential (GWP). HFO-1234ze(E) offers a 99.9% reduction in GWP compared to HFA-134a, aligning with sustainability goals in respiratory care. While it is not an active pharmaceutical ingredient, it serves as the aerosol delivery vehicle for various inhaled medications used to treat asthma and chronic obstructive pulmonary disease (COPD). Clinical development programs are currently assessing the safety, tolerability, and bioequivalence of HFO-1234ze(E)-based formulations, including its impact on mucociliary clearance in healthy participants.
