hgp100 + ATCV is an allogeneic whole-cell tumor vaccine immunotherapy that consists of an allogeneic tumor cell vaccine engineered to express the human glycoprotein 100 (hgp100), a melanoma-associated differentiation antigen. The purpose of incorporating xenogeneic (from another species, in this case human) gp100 into the vaccine is to break immune tolerance and provoke a stronger anti-tumor immune response. This approach allows cross-reaction between the xenogeneic antigen and host molecules, theoretically enabling the immune system to target tumor-associated antigens and induce clinically relevant tumor regression. The vaccine is administered intradermally and has been studied primarily for the treatment of naturally occurring melanoma in pet dogs. Clinical studies demonstrated that the vaccine was well tolerated, with an overall response rate (complete and partial responses) of 17% and a tumor control rate (including stable disease) of 35%. The vaccine elicited anti-hgp100 antibodies and enhanced immune responses, though these did not always correlate with clinical outcomes.
