Preclinical-stage antisense oligonucleotide being developed by OliPass Corporation that targets hypoxia-inducible factor-1 alpha (HIF-1α) for the treatment of solid tumors as a chemo/radio-sensitizer. The drug uses OliPass's proprietary OPNA (OliPass Peptide Nucleic Acids) technology, which consists of peptide nucleic acids (PNA) that have been chemically modified with cationic moieties to enhance cell permeability and stability while maintaining high binding affinity to target nucleic acids. The mechanism of action involves an exon-skipping approach where the PNA antisense oligonucleotide induces skipping of exon 2 in HIF-1α pre-mRNA, resulting in reduced expression of full-length HIF-1α mRNA and protein. HIF-1α is a master regulator of cellular response to hypoxia and is frequently overexpressed in solid tumors, making it an attractive target for cancer therapy. The compound is designed to be delivered as a 'naked' oligonucleotide without requiring additional formulation aids, and is intended to sensitize tumors to chemotherapy and radiation by inhibiting hypoxia-induced survival pathways.
