Overview

The HIV-1 gag DNA vaccine (Wyeth) is an experimental DNA-based biologic therapeutic developed by Wyeth (now Pfizer) for the treatment of HIV-1 infection. The vaccine utilizes a plasmid DNA platform encoding an RNA-optimized truncated version of the HIV-1 gag gene (p37), derived from the HXB2 strain. It is formulated with bupivacaine, which acts as a simple chemical adjuvant to enhance DNA uptake. In Phase 1 clinical trials (such as HVTN 060 and HVTN 063), the vaccine was evaluated both alone and in combination with molecular adjuvants—plasmid DNAs encoding human cytokines IL-12 or IL-15—to boost cellular immune responses. The primary objective of this therapeutic approach is to stimulate or enhance HIV-specific T-cell responses in HIV-infected individuals, particularly those on stable highly active antiretroviral therapy (HAART).
