Overview

HIV glycoprotein 140 + LTK63 is an experimental HIV-1 vaccine candidate developed by St George's, University of London. The vaccine consists of a recombinant, soluble form of the HIV-1 envelope glycoprotein (gp140), which represents the ectodomain of the viral spike protein (often with the V2 loop deleted), combined with LTK63, a genetically detoxified mutant of the Escherichia coli heat-labile toxin (LT) used as a mucosal adjuvant. The candidate was evaluated in a Phase I clinical trial for its ability to induce mucosal and systemic immune responses via intranasal and intramuscular administration (the latter using MF59 adjuvant). However, the development was halted after the trial was terminated early due to safety concerns, specifically the occurrence of transient facial nerve paralysis (Bell's palsy) in participants who received the intranasal formulation containing the LTK63 adjuvant.
