The HIV-MAG DNA vaccine regimen is an investigational preventive HIV vaccine strategy developed through a collaboration between Ichor Medical Systems, the International AIDS Vaccine Initiative (IAVI), and Profectus Biosciences. The regimen utilizes a prime-boost approach centered on a multi-antigenic plasmid DNA (pDNA) vaccine (HIV-MAG) that encodes several HIV-1 proteins, including Gag, Pol, Nef, Tat, Vif, and Envelope. To enhance the resulting immune response, the DNA vaccine is co-administered with GENEVAX IL-12, a plasmid DNA encoding interleukin-12 that acts as a molecular adjuvant. Delivery of these DNA components is achieved via intramuscular injection followed by electroporation using Ichor Medical Systems' proprietary TriGrid Delivery System (TDS-IM), which significantly increases cellular uptake. The protocol may also include a subsequent boost with recombinant adenovirus serotype 35 (Ad35) viral vectors (Ad35-GRIN/Env) to further stimulate robust CD4+ and CD8+ T-cell mediated immunity against HIV antigens.
