Overview

This is an investigational multi-peptide vaccine designed for the treatment of pediatric gliomas. It consists of HLA-A2-restricted synthetic epitope peptides derived from three glioma-associated antigens (GAA): EphA2, interleukin-13 receptor alpha 2 (IL-13Rα2), and survivin. The vaccine is emulsified in the adjuvant Montanide-ISA-51 and co-administered with polyinosinic-polycytidylic acid stabilized by lysine and carboxymethylcellulose (poly-ICLC), a Toll-like receptor 3 (TLR3) agonist. The mechanism of action involves the induction of a systemic cytotoxic T lymphocyte (CTL) response specifically targeting tumor cells that overexpress these antigens. It has been evaluated in pilot studies for children and young adults with newly diagnosed or recurrent high-grade and low-grade gliomas, including diffuse intrinsic pontine glioma (DIPG).
