Overview

HLA-A2 restricted synthetic tumor antigen vaccine + imiquimod is an investigational peptide-based cancer vaccine regimen developed by James Felker at the University of Pittsburgh. The therapy consists of synthetic peptides representing tumor-associated antigens (TAAs) that are specifically presented by the Human Leukocyte Antigen (HLA)-A2 allele. This vaccine is designed to treat children and young adults with recurrent ependymoma by inducing a cytotoxic T-cell response against the tumor cells. To enhance the immune response, the vaccine is administered subcutaneously in conjunction with imiquimod, a topical Toll-like receptor 7 (TLR7) agonist that acts as an immunoadjuvant. The treatment is currently being evaluated in Phase 1 clinical trials to assess safety, tolerability, and immunogenicity in HLA-A2 positive patients.
