HLX07 is a novel, humanized anti-Epidermal growth factor receptor (EGFR) monoclonal antibody developed by Henlius. It was engineered by re-humanizing the Fab regions and minimizing glycan content to reduce immunogenicity while maintaining high affinity for EGFR, aiming to improve upon cetuximab. Preclinical and clinical studies have shown that HLX07 binds EGFR with similar or better bioactivity compared to cetuximab and can inhibit tumor growth in various models. When combined with chemotherapy agents such as gemcitabine (a nucleoside analog inhibiting DNA synthesis) and cisplatin (a platinum-based DNA crosslinker), HLX07 has demonstrated safety, tolerability, and preliminary efficacy in patients with advanced solid tumors[1][3][5]. The combination is being investigated primarily for advanced solid tumors including esophageal squamous cell carcinoma (ESCC) and cutaneous squamous cell carcinoma (CSCC)[2][4].
