**HLX208 + serplulimab** is an unbranded investigational combination consisting of HLX208, a small molecule inhibitor of BRAF V600E, and serplulimab (HLX10, branded as HANSIZHUANG), a humanized monoclonal IgG4 antibody targeting programmed cell death protein 1 (PD-1). HLX208 selectively inhibits mutant BRAF kinase activity, blocking the MAPK signaling pathway and resulting in antitumor effects in BRAF V600E-mutated cancers. Serplulimab acts as an immune checkpoint inhibitor by blocking PD-1, enhancing T-cell mediated antitumor immunity. These agents are being developed by Shanghai Henlius Biotech and have shown antitumor activity, both individually and in combination regimens, primarily targeting cancers with BRAF mutations and malignancies treatable by PD-1 blockade.
