HLX22 + trastuzumab + nab-paclitaxel + gemcitabine is a combination therapy being investigated for the first-line treatment of HER2-positive metastatic pancreatic ductal adenocarcinoma. HLX22 is a recombinant humanized monoclonal antibody that targets the extracellular subdomain IV of the human epidermal growth factor receptor 2 (HER2) at an epitope distinct from that of trastuzumab. This allows both antibodies to bind simultaneously to HER2, leading to a dual blockade that synergistically inhibits HER2 signaling and promotes receptor internalization and degradation. The addition of this dual HER2-targeted approach to the standard-of-care chemotherapy regimen of gemcitabine and nab-paclitaxel (GEM-NABP) aims to improve clinical outcomes in patients with HER2-expressing pancreatic malignancies, a population with high unmet medical needs.
