HLX43 + HLX07 is a combination therapy developed by Shanghai Henlius Biotech for the treatment of advanced or metastatic colorectal cancer. HLX43 is a humanized anti-PD-L1 antibody-drug conjugate (ADC) that utilizes a novel DNA topoisomerase I (topo I) inhibitor payload with a drug-antibody ratio (DAR) of 8. It is designed to provide dual anti-tumor activity through immune checkpoint blockade and payload-mediated cytotoxicity. HLX07 is a recombinant humanized monoclonal antibody targeting the epidermal growth factor receptor (EGFR), engineered for lower immunogenicity and higher target affinity compared to cetuximab. This combination is currently being evaluated in Phase 1b/2 clinical trials for patients who have failed or are intolerant to standard first-line therapies.
