HLX43 + HLX10 + HLX07 is a combination therapy developed by Shanghai Henlius Biotech for the treatment of recurrent or metastatic head and neck squamous cell carcinoma (HNSCC). The regimen consists of HLX43, a potential best-in-class antibody-drug conjugate (ADC) targeting PD-L1; HLX10 (serplulimab), a humanized monoclonal antibody targeting the PD-1 immune checkpoint; and HLX07, a recombinant humanized monoclonal antibody targeting the epidermal growth factor receptor (EGFR). HLX43 utilizes MediLink Therapeutics' TMALIN® technology to conjugate a topoisomerase I inhibitor payload to an anti-PD-L1 antibody with a drug-antibody ratio (DAR) of 8. This triple combination aims to leverage the synergistic effects of immune checkpoint blockade (PD-1/PD-L1) and EGFR inhibition alongside payload-mediated cytotoxicity to overcome resistance and improve outcomes in HNSCC.
