HLX43 + serplulimab is an investigational combination therapy currently in clinical development for advanced/metastatic solid tumors. **HLX43** is an antibody-drug conjugate (ADC) composed of a fully humanized anti-PD-L1 IgG1 antibody linked via a novel tripeptide linker to a topoisomerase inhibitor payload. It acts via dual mechanisms: (1) targeted cytotoxic delivery, inducing tumor cell apoptosis by delivering the cytotoxic payload to PD-L1 expressing tumor cells, and (2) immune checkpoint blockade via PD-L1/PD-1 axis inhibition. **Serplulimab** is a humanized monoclonal IgG4 antibody targeting PD-1, acting as an immune checkpoint inhibitor to restore tumor-specific T-cell response by blocking interaction of PD-1 with its ligands, PD-L1 and PD-L2. The combination seeks to exploit synergy between ADC-mediated cytotoxicity and PD-1 axis immune activation. It is being evaluated in phase 1/2 trials in advanced solid tumors, including non-small cell lung cancer, thymic squamous cell carcinoma, hepatocellular carcinoma, esophageal squamous cell carcinoma, head and neck squamous cell carcinoma, cervical cancer, and nasopharyngeal carcinoma[1][3][4][7].
