HLX87 + HLX22 is a combination therapy being developed by Shanghai Henlius Biotech for the treatment of HER2-positive breast cancer. The combination consists of HLX22, a novel-epitope recombinant humanized anti-HER2 monoclonal antibody, and HLX87, a HER2-targeted antibody-drug conjugate (ADC). HLX22 binds to subdomain IV of the HER2 extracellular region at an epitope distinct from trastuzumab, which promotes receptor internalization and degradation. HLX87 comprises an anti-HER2 antibody conjugated to a topoisomerase I inhibitor payload via a stable, cleavable linker, delivering cytotoxic activity directly to HER2-expressing cells. This dual-targeting approach is designed to enhance anti-tumor efficacy in patients with HER2-positive recurrent or metastatic breast cancer and is currently being evaluated in Phase 2/3 clinical trials.
