HLX87 + pertuzumab is a combination therapy being developed by Shanghai Henlius Biotech for the treatment of HER2-positive breast cancer. HLX87 is an antibody-drug conjugate (ADC) consisting of a humanized anti-HER2 monoclonal antibody conjugated to a topoisomerase I inhibitor payload via a stable, cleavable open-chain linker. Pertuzumab is a recombinant humanized monoclonal antibody that targets the extracellular dimerization domain (subdomain II) of the human epidermal growth factor receptor 2 (HER2). The combination is designed to provide dual HER2 blockade and potent cytotoxic delivery to overcome resistance and improve efficacy in patients with recurrent or metastatic disease. It is currently being evaluated in a Phase 2/3 clinical trial (HLX87-BC001) as a first-line treatment.
