Overview

HM1.24-ETA' is an experimental, preclinical recombinant single-chain immunotoxin designed for the treatment of CD317-expressing malignancies, including multiple myeloma, plasma cell leukemia, and glioblastoma. It is a biologic fusion protein consisting of a CD317-specific (HM1.24) single-chain variable fragment (scFv) antibody genetically fused to ETA', a truncated form of Pseudomonas aeruginosa exotoxin A that lacks the cell-binding domain I. HM1.24-ETA' targets CD317 (bone marrow stromal antigen 2 / BST2), which is overexpressed on malignant plasma cells and glioma cells. Upon binding, the immunotoxin is internalized, and the ETA' payload inhibits protein synthesis, leading to cell death via apoptosis. The recombinant protein is expressed in Escherichia coli. HM1.24-ETA' was developed by researchers at Kiel University and has been evaluated in collaboration with the University of Zurich.
