HMBD-001 + docetaxel is a combination investigational therapy composed of HMBD-001, a humanized IgG1 monoclonal antibody targeting HER3 (ERBB-3), and docetaxel, a chemotherapeutic agent of the taxane class. HMBD-001 binds to a unique epitope at the domain II dimerization interface of HER3, effectively blocking EGFR/HER2-HER3 heterodimerization and potently inhibiting PI3K signaling, irrespective of ligand binding to the receptor. This mechanism prevents HER3-driven tumor growth and drug resistance. Docetaxel stabilizes microtubules, preventing cell division and inducing apoptosis in proliferating cancer cells. The combination is being explored in solid tumors, particularly those with HER3 signaling aberrations such as NRG1 fusions or HER3 mutations, including squamous cell carcinoma and other HER3-driven cancers in advanced/metastatic settings.
