HMPL-523 + azacitidine is an investigational combination therapy consisting of **HMPL-523**, a selective spleen tyrosine kinase (Syk) inhibitor, and **azacitidine**, a nucleoside metabolic inhibitor that is approved for certain hematologic malignancies. HMPL-523 was developed to block Syk-mediated signaling involved in cancer cell survival, proliferation, and immune modulation in hematological cancers. Azacitidine works by inhibiting DNA methylation, thus restoring normal gene function and promoting cancer cell death. The combination is being evaluated primarily in elderly patients with previously untreated acute myeloid leukemia (AML) who are ineligible for standard induction therapy. This combination is under clinical investigation by Hutchison China MediTech (Hutchmed) in phase I clinical trials in China, focusing on safety, pharmacokinetics, and preliminary efficacy[1].
