The HAD regimen is a combination chemotherapy protocol consisting of homoharringtonine (HHT), cytarabine (Ara-C), and daunorubicin (DNR). It is primarily being investigated by the Institute of Hematology & Blood Diseases Hospital, Chinese Academy of Medical Sciences, for the treatment of acute myeloid leukemia (AML), with a specific focus on patients harboring CEBPA double mutations (CEBPAdm). Homoharringtonine is a plant alkaloid that inhibits protein synthesis by binding to the ribosome; cytarabine is a nucleoside analog that interferes with DNA synthesis by inhibiting DNA polymerase; and daunorubicin is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II. This combination therapy targets rapidly dividing leukemic cells through multiple synergistic mechanisms and is being compared against the standard 3+7 induction regimen in clinical trials.
