Overview

HPP071 (also known as HPP971 or ANT-401) and HPP3033 are non-electrophilic small molecule modulators of the Nrf2/Bach1 pathway developed by vTv Therapeutics. These compounds function by inhibiting Bach1, a transcriptional repressor that competes with Nrf2 for binding to antioxidant response elements (ARE) in the promoter regions of various genes. By inhibiting Bach1, these molecules facilitate the activation of the Nrf2 (Nuclear factor erythroid 2-related factor 2) pathway, which induces the expression of antioxidant and anti-inflammatory genes to mitigate oxidative stress and inflammation. HPP971 has completed Phase 1 single and multiple ascending dose studies and was licensed to Anteris Biopharma for development in renal indications, while HPP3033 is a preclinical development candidate. The program targets a variety of conditions, including renal injury, Parkinson's disease, mitochondrial myopathies, and non-alcoholic steatohepatitis (NASH).
