This combination therapy regimen is being developed by Shanghai Hengrui Pharmaceutical Co., Ltd. for the treatment of pancreatic adenocarcinoma, specifically targeting patients with KRAS G12D mutations. The regimen integrates four distinct therapeutic agents: HRS-4642, a highly selective small molecule inhibitor of the KRAS G12D mutation; adebrelimab (also known as SHR-1316), a humanized IgG4 monoclonal antibody that acts as a PD-L1 checkpoint inhibitor; and the standard chemotherapy doublet of nab-paclitaxel (albumin-bound paclitaxel) and gemcitabine. By combining targeted inhibition of a primary oncogenic driver with immunotherapy and cytotoxic chemotherapy, the regimen aims to achieve synergistic anti-tumor activity. It is currently under evaluation in Phase 1b/2 clinical trials for locally advanced or metastatic pancreatic cancer.
