Overview

HRS-4642 + chemotherapy + adebrelimab is an investigational combination regimen being evaluated for the treatment of KRAS G12D-mutant cancers, particularly pancreatic ductal adenocarcinoma (PDAC). The regimen consists of HRS-4642, a selective, non-covalent KRAS G12D inhibitor; a chemotherapy backbone (typically gemcitabine and albumin-bound paclitaxel, or platinum-based doublets); and adebrelimab (SHR-1316), a humanized IgG4 monoclonal antibody targeting PD-L1. Developed by Jiangsu Hengrui Pharmaceuticals, this combination aims to synergistically target the oncogenic driver KRAS G12D while simultaneously reversing tumor-induced immunosuppression via PD-L1 blockade and enhancing cytotoxic tumor cell death through chemotherapy. It is currently in Phase 2 clinical trials for pancreatic cancer.
