HRS-4642 + nab-paclitaxel + gemcitabine is an investigational combination therapy developed by Shanghai Hengrui Pharmaceutical Co., Ltd. for the treatment of advanced or metastatic pancreatic adenocarcinoma harboring the KRAS G12D mutation. This regimen combines HRS-4642, a potent and highly selective small molecule inhibitor of the KRAS G12D mutant protein, with the standard cytotoxic chemotherapy backbone of nab-paclitaxel (an albumin-bound formulation of paclitaxel that inhibits microtubule disassembly) and gemcitabine (a nucleoside analog that inhibits DNA synthesis). By targeting the most common oncogenic driver in pancreatic cancer alongside traditional chemotherapy, the combination aims to provide synergistic anti-tumor activity and overcome resistance mechanisms. It is currently being evaluated in Phase Ib/II clinical trials, both as a triplet and in combination with immunotherapy.
