HRS-7058 + SHR-9839 is an investigational combination therapy being developed by Shandong Suncadia Medicine Co., Ltd., a subsidiary of Jiangsu Hengrui Pharma, for the treatment of advanced malignant solid tumors, particularly those harboring KRAS G12C mutations. HRS-7058 is a small molecule inhibitor that specifically and covalently binds to the KRAS G12C mutant protein, locking it in an inactive GDP-bound state and thereby suppressing downstream oncogenic signaling pathways such as MAPK/ERK. SHR-9839 is a monoclonal antibody targeting T-cell immunoreceptor with Ig and ITIM domains (TIGIT), an inhibitory immune checkpoint receptor expressed on T cells and natural killer (NK) cells. The combination of a KRAS G12C inhibitor with a TIGIT antagonist is designed to provide a synergistic effect by directly inhibiting tumor cell proliferation and simultaneously overcoming immune evasion, potentially improving clinical outcomes in patients with KRAS-mutated solid tumors.
