Overview

HS-10370 + pemetrexed + cisplatin is a combination regimen under clinical investigation for the treatment of advanced solid tumors, especially non-small cell lung cancer (NSCLC) with KRAS G12C mutations. HS-10370 is a potent, highly-selective, orally-administered small molecule inhibitor of KRAS G12C, developed by Hansoh Pharma, that inhibits the aberrant activity of the mutant KRAS protein, thereby blocking downstream signaling pathways and inhibiting cancer cell proliferation. Pemetrexed is an antimetabolite chemotherapeutic agent that disrupts folate-dependent metabolic processes essential for cell replication, while cisplatin is a platinum-based chemotherapeutic that causes DNA crosslinking and apoptosis in cancer cells. In combination, these agents are being evaluated for synergistic activity and superior efficacy in patients with solid tumors harboring actionable KRAS mutations, primarily NSCLC, where there remains a significant unmet medical need[1][3][5][7][8].
