HS-10382 + flumatinib is a combination therapy under clinical investigation for the treatment of chronic myeloid leukemia (CML), particularly in patients resistant or intolerant to previous tyrosine kinase inhibitors (TKIs). HS-10382 (TERN-701) is a potent, orally bioavailable small molecule allosteric inhibitor of BCR-ABL1 that acts by binding to the myristoyl site of the BCR-ABL1 protein, locking it in an inactive conformation and offering a unique resistance profile compared to ATP-binding site TKIs. Flumatinib is a second-generation BCR-ABL1 tyrosine kinase inhibitor already approved in China as an imatinib derivative. This combination aims to target both distinct sites on BCR-ABL1 to overcome resistance mechanisms to single-agent TKI therapies[1][3][5].
