HS628 is a proposed biosimilar of tocilizumab, a recombinant humanized monoclonal antibody that targets the interleukin-6 receptor (IL-6R). Tocilizumab inhibits IL-6-mediated signal transduction by blocking IL-6 binding to both soluble and membrane-bound forms of its receptor. It is primarily indicated for rheumatoid arthritis and juvenile idiopathic arthritis. Methotrexate is an antimetabolite and antifolate agent used in the treatment of various cancers, rheumatoid arthritis, psoriasis, and other inflammatory conditions. Its mechanism involves inhibition of enzymes responsible for nucleotide synthesis—most notably dihydrofolate reductase—thereby preventing cell division and exerting anti-inflammatory effects. The combination of an IL-6R inhibitor with methotrexate is commonly used in clinical practice for enhanced efficacy in autoimmune diseases such as rheumatoid arthritis[5][6][7].
