HSK31858 + rifampicin is a combination drug made up of HSK31858, a potent, oral small molecule inhibitor of dipeptidyl peptidase-1 (DPP-1, also known as cathepsin C), and rifampicin, an established antibiotic primarily used to treat tuberculosis and other bacterial infections. HSK31858 acts by reversibly inhibiting DPP-1, leading to reduced activation of neutrophil serine proteases and potentially lowering inflammation in airway diseases[1][2][3][4][6][7][8]. Rifampicin is a broad-spectrum antibiotic that inhibits bacterial DNA-dependent RNA polymerase, suppressing bacterial RNA synthesis. The combination may be used in clinical pharmacokinetic or drug interaction studies (such as to assess the impact of enzyme induction on HSK31858 metabolism), but is not an established therapeutic product with recognized respiratory indications. HSK31858 alone has been developed by Haisco Pharmaceutical and licensed to Chiesi for global development, showing efficacy in reducing bronchiectasis exacerbations by targeting neutrophilic airway inflammation[1][3][4][6][7][8].
