Overview

**HSV(IE)Aβ(CMV)IL-4** is an experimental herpes simplex virus amplicon-based Alzheimer's disease vaccine developed at the University of Rochester. The nonreplicating HSV vector co-delivers an amyloid beta peptide 1-42 antigen cassette and an interleukin-4 expression cassette, aiming to generate an anti-amyloid beta antibody response biased toward a Th2-like immune profile while limiting potentially harmful inflammatory responses. In triple-transgenic Alzheimer's disease mice, subcutaneous vaccination was associated with increased Th2-related amyloid beta-specific antibodies, improved learning and memory performance, and reduced amyloid and tau pathology.
