A combination immunotherapy regimen for the treatment of HIV-1 infection, consisting of the HTI (HIVACAT T-cell immunogen) therapeutic vaccine and the TLR7 agonist vesatolimod (GS-9620). The HTI vaccine is delivered using a prime-boost strategy with recombinant viral vectors—Chimpanzee Adenovirus Oxford 1 (ChAdOx1) and Modified Vaccinia Virus Ankara (MVA)—to elicit a broad and potent T-cell response against conserved regions of the HIV genome. Vesatolimod is an orally administered small molecule that selectively activates Toll-like receptor 7, thereby stimulating innate immune pathways and potentially enhancing the vaccine-induced T-cell response or promoting the clearance of virally infected cells from the reservoir. This combination was developed through a collaboration between Aelix Therapeutics and Gilead Sciences.
