This experimental cell therapy consists of human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) that have been pre-activated or "primed" with the pro-inflammatory cytokine interferon-gamma (IFN-gamma). Developed by the Research Institute of Surgery at Daping Hospital (Third Military Medical University), this approach aims to enhance the therapeutic potential of MSCs for autoimmune conditions like rheumatoid arthritis. Priming with IFN-gamma "licenses" the MSCs, significantly upregulating the expression of indoleamine 2,3-dioxygenase (IDO), an enzyme that plays a critical role in immunosuppression by depleting tryptophan and producing kynurenines. This mechanism leads to the inhibition of T-cell proliferation and the modulation of the Th17/Treg balance, shifting the immune environment from a pro-inflammatory state toward immune tolerance. The therapy is typically administered systemically to reduce joint inflammation and prevent bone erosion in patients with rheumatoid arthritis.
