**Humanized GPC1-ADC (MMAE)** is an academic, preclinical glypican-1-targeted antibody-drug conjugate composed of the humanized anti-GPC1 monoclonal antibody clone T2 conjugated to monomethyl auristatin E through maleimidocaproyl-valine-citrulline-p-aminobenzyloxycarbonyl cleavable linkers. It binds cell-surface glypican-1, is internalized by GPC1-positive tumor cells, and releases MMAE intracellularly, causing microtubule disruption, G2/M-phase arrest, and apoptosis. The vc-PABC linker and membrane-permeable MMAE payload also support bystander killing of adjacent GPC1-negative tumor cells. The construct has shown preclinical activity in GPC1-positive pancreatic ductal adenocarcinoma, esophageal squamous cell carcinoma, glioblastoma, gastric cancer, and lung squamous cell carcinoma models; no INN, commercial brand, clinical trial program, regulatory approval, or commercial manufacturer has been identified.
