This is a third-generation CAR-T cell therapy targeting the interleukin-13 receptor alpha 2 (IL-13Rα2) for the treatment of glioblastoma (GBM). Developed by researchers at the Clinical Center of Gene and Cell Engineering, Beijing Shijitan Hospital, Capital Medical University, the therapy utilizes a humanized single-chain variable fragment (scFv) as the antigen-binding domain to minimize potential immunogenicity compared to murine-derived binders. The CAR construct incorporates both CD28 and 4-1BB co-stimulatory domains alongside the CD3ζ signaling domain and a CD28 transmembrane domain. Preclinical studies published in 2022 demonstrated that these humanized CAR-T cells exhibit superior tumor-killing effects, expansion, and persistence compared to murine counterparts in both subcutaneous and intracranial glioblastoma mouse models.
