HX008 + pemetrexed + cisplatin + carboplatin is an investigational multidrug combination consisting of HX008, a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), with three chemotherapeutic agents: pemetrexed, cisplatin, and carboplatin. HX008 blocks PD-1, disrupting PD-1/PD-L1 and PD-1/PD-L2 interactions and thereby reactivating tumor-infiltrating lymphocytes and promoting immune-mediated tumor cell killing. Pemetrexed is an antimetabolite that inhibits thymidylate synthase, dihydrofolate reductase, and glycinamide ribonucleotide formyl transferase; cisplatin and carboplatin are platinum-based agents that cause DNA crosslinking, leading to apoptosis of rapidly dividing tumor cells. The combination is studied for enhanced efficacy in treating advanced or metastatic non-small cell lung cancer (NSCLC) and potentially other advanced solid tumors, aiming to combine immune checkpoint blockade with cytotoxic chemotherapy for synergistic antitumor effects[1][3][4][6].
