**HA + gemcitabine** (also known as hyaluronan-gemcitabine or HyACT-gemcitabine) is a tumor-targeted formulation of the chemotherapeutic drug gemcitabine combined with hyaluronic acid (HA). Developed primarily under Alchemia's proprietary HyACT® (Hyaluronic Acid ChemoTransport) platform, this formulation utilizes HA as a targeting vehicle to selectively deliver gemcitabine to CD44-overexpressing cancer cells, including cancer stem cells (CSCs). CD44 is a cell-surface glycoprotein that is highly expressed on various solid tumors and serves as a key marker for CSCs, which are often resistant to standard chemotherapy. By binding to CD44, the HA-gemcitabine formulation is internalized via receptor-mediated endocytosis, enhancing intracellular drug accumulation and cytotoxicity while potentially reducing systemic side effects. Preclinical studies have demonstrated its efficacy in breast cancer, pancreatic cancer, glioblastoma, and lung cancer models.
