Overview

**Hydralazine + valproic acid** is an investigational oral epigenetic anticancer combination administered as two separate small-molecule drugs. Hydralazine has been studied as a DNA hypomethylating agent, while valproic acid inhibits histone deacetylases. Together, the regimen is intended to reverse epigenetic silencing of tumor-suppressor, antigen-presentation, and other growth-regulatory genes, potentially enhancing tumor immunogenicity and sensitivity to chemotherapy and radiotherapy. The combination has been evaluated in cervical cancer, breast cancer, advanced hepatocellular carcinoma, myelodysplastic syndromes, refractory solid tumors, and advanced previously treated solid tumors. It is not an approved fixed-dose combination product.
