Overview

Hydroxychloroquine + atorvastatin is a combination of two small molecule drugs, hydroxychloroquine and atorvastatin. Hydroxychloroquine is an antimalarial and disease-modifying anti-rheumatic drug (DMARD) that acts by interfering with lysosomal activity and antigen presentation in immune cells, as well as modulating inflammatory pathways. Atorvastatin is a selective, competitive inhibitor of HMG-CoA reductase, the rate-limiting enzyme in cholesterol biosynthesis, leading to reduced cholesterol levels and pleiotropic effects such as improved endothelial function and anti-inflammatory actions. The combination has been studied for its potential synergistic effects on glycemic control and lipid profile improvement in diabetes mellitus models[1], as well as for reducing inflammation and pain in osteoarthritis[2]. Both drugs are FDA-approved individually but the fixed-dose combination does not have a unique brand name or specific regulatory approval.
