A combination therapy consisting of hydroxychloroquine, metformin, and methotrexate. This combination has been used in specific clinical contexts, particularly for autoimmune and inflammatory conditions. Each component has distinct mechanisms of action: hydroxychloroquine is an antimalarial with immunomodulatory effects that inhibits autophagy; metformin is an antidiabetic medication with anti-inflammatory properties; and methotrexate is an antifolate drug used for inflammatory conditions and cancer. Important drug interactions exist between these medications, including increased risk of fatal outcomes with hydroxychloroquine and metformin combination, and potential for increased methotrexate toxicity when combined with hydroxychloroquine.
